Site-specific recombinase (SSR) system is widely used in genetically modification, especially the generation for conditional KO and inducible KO mouse models. For making it more efficient and easier, mouse models with special functions have been generated, such as tissue-specifically expressing Cre, inducible expressing Cre, Split-Cre, floxed (target gene were flanked by loxP sites) mouse, et al.

The ArcCreER knock-in/knockout allele was designed to both abolish expression of the Arc immediate early gene and direct expression of the CreERT2 fusion protein from the Arc promoter/enhancer elements. These mice are a Cre-lox tool that allows inducible Cre expression in Arc-expressing cells/tissues; including neurons activated by specific somatosensory, visual, and auditory stimuli. Immediate early genes (IEGs) are the connection between gene expression and a neuron's electrical/synaptic activity (which defines the neuron's response properties).

Fos2A-iCreER knock-in (also called Fos2A-iCreERT2 or "TRAP2") was designed to have expression of a tamoxifen-inducible, improved Cre recombinase (icre/ERT2) from the Fos promoter/enhancer elements - without disrupting endogenous Fos expression. Fos2A-iCreER mice are a Cre-lox tool allowing inducible iCre recombination in Fos-expressing cells/tissues (including neurons activated by specific somatosensory, visual and auditory stimuli) or behavioral experiences such as enriched environment, fear conditioning or water deprivation. ROSAnT-nG mice are available on a congenic C57BL/6NJ background and the congenic version will replace this mixed background strain in the near future. ROSAnT-nG (also called nT/nG) is a nuclear-targeted, two-color fluorescent Cre reporter allele that express nuclear-localized red fluorescence in a widespread fashion prior to Cre recombinase exposure. Following exposure to Cre, nuclear-localized green fluorescence is observed. In addition to this B6;129 mixed genetic background ROSAnT-nG strain, the ROSAnT-nG reporter allele is also available on a C57BL/6NJ congenic background. The ROSAnT-nG reporter allele functions similarly to the mT/mG reporter allele (ROSAmT/mG), with nT/nG directed to nuclei and mT/mG directed to cell membrane.

The Snap25-LSL-2A-EGFP-D (also called Snap25-LSL-2A-EGFP-Δ or Snap25-LSL-2A-EGFP-Δneo/hygro) conditional allele is designed with a loxP-flanked STOP cassette and enhanced green fluorescent protein (EGFP) downstream of the synaptosomal-associated protein 25 (Snap25) translational STOP codon. Following Cre recombinase removal of the floxed STOP cassette, EGFP fluorescence is expected in cells/tissues where the expression patterns of Snap25 and Cre recombinase overlap. Snap25 is a t-SNARE protein. These mice may be useful as a Cre-lox tool for fluorescent visualization of synaptic vesicle/plasma membrane fusion.
