Introduction into Brainbow
The brainbow technology is based on site-specific recombinase system, resulting in the expression of multiple fluorescent proteins in a single cell. It is a genetic technology used to label and visualize individual cells with in tissues or organs, especially in the nervous system. This technology was first applied in mice, then adapted in other model organisms such as zebrafish, fruit flies and C. elegans.
The Brainbow technique has been further developed and optimized since its initial creation. Variants of the technique include Confetti, allowing for clonal labeling, and Brainbow 3.0, 3.1 and 3.2, which allow for greater spectral diversity and improved expression levels.
Strategies of brainbow (Cai, Dawen et al., 2013)
Brainbow works by introducing multiple copies of a gene encoding for fluorescent proteins, such as green, red, and blue, into a group of cells. The copies of the gene are randomly recombined using site-specific recombination enzymes, resulting in each cell expressing a unique combination of fluorescent proteins. This allows researchers to differentiate individual cells based on their unique color signature.
