Introduction into Xenopus Models Generated by T-SSR System

One of the advantages of using Xenopus as a model organism for site-specific recombinase technology is their external fertilization and transparent embryos, which allow for easy access to specific cells and tissues for targeted gene modifications. In addition, the large size of Xenopus embryos and their ability to survive without a functional cardiovascular system during early development make them suitable for genetic manipulations that may not be feasible in other model organisms.

In Xenopus, researchers have used Cre/loxP technology to generate human disease models, such as cancer and neurological disorders. In these models, the Cre recombinase is expressed under the control of a tissue-specific promoter, allowing for conditional gene knockout or overexpression in that tissue or cell type. These models have been valuable in elucidating the molecular mechanisms underlying these diseases.

Another example is the use of T-SSR technology to study the role of the homeobox transcription factor Otx2 in eye development. By introducing loxP sites into the Otx2 gene and crossing these frogs with a transgenic line expressing Cre recombinase under the control of an eye-specific promoter, researchers were able to selectively delete Otx2 in the developing eye and observe its effects on eye development.
