Site-specific recombinase (SSR) system is widely used in genetically modification, especially the generation for conditional KO and inducible KO mouse models. For making it more efficient and easier, mouse models with special functions have been generated, such as tissue-specifically expressing Cre, inducible expressing Cre, Split-Cre, floxed (target gene were flanked by loxP sites) mouse, et al.

Ai161(TIT2L-GFP-ICR-tTA2)-D (also called Ai161D) mice are a Dre and Cre-dependent, Tet-controllable fluorescent reporter line, created by targeted insertion at the Igs7 locus (TIGRE; an intergenic region on mouse chromosome 9 that allows reporter expression to be tightly regulated). Exposure to Dre recombinase removes the rox-flanked STOP cassette and results in tTA2 expression. Subsequent exposure to Cre recombinase removes the loxP-flanked STOP cassette - allowing the tTA2 to induce EGFP expression. The EGFP expression may then be diminished by doxycycline. Ai161D mice allow intersectional genetic fate mapping of different cell subpopulations defined by the overlap of the two gene promoters driving the expression of each recombinase.

LoxCode knock-in mice have the LoxCode cassette inserted into the endogenous Gt(ROSA)26Sor locus. The LoxCode cassette is composed of 14 loxP sites in alternating orientation flanking 13 small (8-14nts) code elements. Upon Cre exposure, random recombination between the loxP sites lead to inversions and excisions, resulting in DNA sequence reshuffling with a theoretical DNA diversity of over 30 billion barcodes. This is an inducible, cellular barcoding technology based on the Cre-loxP system that allows for sensitive, high diversity lineage tracing at the cellular, spatial, and tissue level.
