Ai35-hVOS1.5 mice have a loxP-flanked STOP cassette that prevents transcription of hVOS1.5 - an optimized hybrid voltage sensor consisting of cerulean fluorescent protein with a C-terminal truncated h-ras membrane-anchoring motif. This Ai35-hVOS1.5 Cre reporter mouse line allows bright fluorescent imaging of rapid voltage changes with high temporal resolution in the cell populations defined by Cre recombinase expression.

The targeting vector is designed with (from 5' to 3') a CMV-IE enhancer/chicken beta-actin/rabbit beta-globin hybrid promoter (CAG), an FRT site, a loxP-flanked STOP cassette (with stop codons in all 3 reading frames and a triple polyA signal), the hVOS1.5 DNA sequence (described below), a woodchuck hepatitis virus post-transcriptional regulatory element (WPRE; to enhance the mRNA transcript stability), a BGH polyA signal and an attB/attP-flanked PGK-FRT Neo-polyA cassette. The entire targeting vector was inserted between exons 1 and 2 of the Gt(ROSA)26Sor locus. hVOS1.5 is an optimized hybrid voltage sensor consisting of cerulean fluorescent protein (CeFP) with a C-terminal truncated h-ras farnesylation motif that tethers it to the inner face of the plasma membrane.

Breeding Considerations
Both heterozygous and homozygous mice are viable and fertile with no reported gross physical or behavioral abnormalities. When maintaining a live colony, heterozygous mice may be bred together or to wildtype mice from the colony. Alternatively, homozygous mice may be bred together.
