These transgenic mice express human CD4 and CCR5 under the regulatory control of the human ubiquitin C promoter in combination with a widely-expressed luciferase reporter regulated by the Gt(ROSA)26Sor promoter. The mice enable the rapid measurement of HIV-1 transmission by peritoneal and genitomucosal HIV-1 pseudovirus entry in vivo. The system represents a model for the pre-clinical evaluation of prophylactic or vaccine candidates.

Molecular Note
A transgene composed of human CCR5 and CD4 on a single poly-protein transcript separated by a ribosomal skip 2A was placed downstream of thehuman ubiquitin C promoter. A linearized 4.5 Kb fragment containing the transgene was injected into C57BL/6J fertilized female pronuclei. Founder line 19 was chosen based on surface expression of hCD4 and hCCR5 on peripheral blood mononuclear cells as determined by flow cytometry.
