The Ant22f (floxed exons 2-3) allele allows Cre-conditional knockout of the adenine nucleotide translocase 2 gene (Slc25a5 or Ant2) on the X chromosome. Removal of the floxed sequence creates a null allele. These mice are useful for spatiotemporal deletions of Ant2 in studying energy metabolism, mitochondrial dysfunction and oxidative stress, as well as cancer or developmental disorders.

Molecular Note
A targeting vector was designed to insert a loxP site upstream of exon 2, and an FRT splice donor/acceptor-IRES-beta-gal/neo-FRT loxP downstream of exon 3. ES cells were transiently transfected with a FLP-expressing plasmid to remove the SA-IRES-betageo cassette leaving exons 2-3 floxed.

Breeding Considerations
The Ant22f allele is on the X chromosome. Homozygous females (Ant22f/2f) and hemizygous males (Ant22f/Y) are viable and fertile with no reported abnormalities. When maintaining a live colony, heterozygous mice may be bred together, to wildtype mice from the colony or to C57BL/6J inbred mice. Alternatively, homozygous females may be bred with hemizygous males.
