The targeting cassette is composed of an FRT site, lacZ sequence, a loxP site, a neomycin resistance gene, a second FRT site, and a second loxP site inserted upstream of exon 4. A third loxP site is inserted downstream of exon 4. FLP-mediated recombination removed the lacZ and neo cassette.
