Tbx18:nlacZ/nGFP knock-in/knock-out has the endogenous T-box18 promoter/enhancer sequences directing expression of nuclear-localized lacZ before exposure to Cre recombinase. Subsequent Cre recombination results in nuclear GFP fluorescence. These mice may be useful in studying pericytes and vascular smooth muscle cells in multiple tissues, as well as proepicardium/epicardium cell lineages, cardiac fibroblasts and pacemaker cells of the sino-atrial node.

A cassette containing a floxed lacZ gene followed by an Hist2h2be-driven EGFP and an FRT flanked neo gene was inserted 6 bp upstream of the endogenous translation initiation codon. The neo cassette was removed by germ line, flp-mediated recombination.

HUSBANDRY
Breeding Considerations
Heterozygous mice are viable and fertile with no reported gross physical or behavioral abnormalities. Homozygous mice are TBX18 null - exhibiting shortened body length and postnatal death due to severe skeletal and respiratory failure. When maintaining a live colony, heterozygous mice may be bred to wildtype littermates.
