Molecular Note
The L1L2_gt0 cassette was inserted upstream of the critical exon (exon 7) with insertion at bp position ChrX:42,222,777 [Build GRCm38]. The L1L2_gt0 cassette is composed of an FRT site, a mouse En2 intron/splice-acceptor/exon (En2 SA), a self-cleaving 2A peptide, the beta-galactosidase gene (lacZ), a second self-cleaving 2A peptide, the neomycin-resistance gene (neo), an SV40 polyadenylation signal (SV40pA), a second FRT site and a loxP site. A second loxP site was inserted downstream of exon 7 at bp position ChrX:42,223,727 [Build GRCm38]; therefore flanking exon 7 with loxP sites. Flp-mediated recombination removed the FRT flanked neo cassette leaving exon 7 floxed.

Breeding Considerations
The floxed allele is on the X chromosome. Homozygous females (Stag2flox/flox) and hemizygous males (Stag2flox/Y) are viable and fertile, with no reported gross phenotypic abnormalities or breeding problems. When maintaining a live colony, homozygous females may be bred to hemizygous males.
