FSF-LSL-SypYFP (or R26-CAG-FSF-LSL-SypYFP) is a dual-recombinase intersectional allele that has a frt-flanked STOP and loxP-flanked STOP that prevent transcription of a synaptophysin-YFP fusion protein. Exposure to FLP recombinase and Cre recombinase results in YFP expression enriched at the synapse/synaptic vesicle (axonal boutons [terminal and en passant]) of the neuron subtype(s) defined by the overlap of FLP/Cre expression.

Molecular Note
The targeting vector was designed with (from 5' to 3') a CMV-IE enhancer/chicken beta-actin/rabbit beta-globin hybrid promoter (from pCAGGS), an FRT flanked STOP cassette (with a short open reading frame, followed by translational stops in all three reading frames, a synthetic polyA sequence, a HGH polyA sequence and a HSV-TK polyA sequence), a loxP-flanked STOP cassette (with stop codons in all three reading frames linked to three repeats of the SV40 polyA signal), the SypYFP fusion gene (a mouse synaptophysin [Syp] cDNA fused to an enhanced yellow fluorescent protein sequence), a woodchuck hepatitis virus post-transcriptional regulatory element, a BGH polyA signal, an AttB site, a PGK promoter-Neomycin resistance gene-PGK pA cassette and an AttP site. This entire targeting vector was inserted between exons 1 and 2 of the Gt(ROSA)26Sor locus.

Breeding Considerations
Mice homozygous for the R26-CAG-FSF-LSL-SypYFP (or FSF-LSL-SypYFP) allele are viable and fertile with normal breeding, and no gross physical or behavioral abnormalities.When maintaining a live colony, heterozygous mice may be bred together, to wildtype mice from the colony or to C57BL/6J inbred animals. Alternatively, homozygous mice may be bred together.
