These α3[5] mice contain a mutation designed to introduce 5 nucleotide mutations to exon 5 of mouse nicotinic acetylcholine receptor (nAChR) neuronal α3 subunit and a loxP-flanked neomycin (neo) resistance cassette upstream of exon 5. These mice may be useful for studying the pathological mechanisms of receptor assembly and trafficking in neurological and psychiatric disorders including Alzheimer's disease, Parkinson's disease, schizophrenia, and epilepsy, as well as drug addiction, response to pain, and nicotine induced behaviors.

Molecular Note
A targeting construct was made containing a mutated exon 5 of mouse nicotinic acetylcholine receptor (nAChR) neuronal alpha3 subunit (alpha3[5]), and a loxP-flanked neomycin (neo) resistance cassette upstream of exon 5. This mutated alpha3[5] gene contained five altered nucleotides, Y184W, E187W, I188V, K189Y, and N191T, in the loop C region corresponding to residues from the alpha1 subunit of nAchR from Torpedo californica, the Pacific electric ray which conveys alpha-bungarotoxin (alphaBTX) sensitivity. Cre mediated recombination removed the neo cassette.
