Fxnflox/null::PV-Cre mice have a Cre-conditional frataxin allele, global knockout frataxin allele and parvalbumin neuron-specific Cre recombinase knock-in allele - useful to study Friedreich's Ataxia.The Fxnflox/null::PV-Cre genotype is compound heterozygous at the frataxin locus (floxed exon 2 and global KO on respective homologous chromosomes) and heterozygous for the PV-Cre knock-in allele. Due to early-onset ataxia, Fxnflox/null::PV-Cre are distributed at 4-7 weeks of age. In addition, we distribute the phenotypically-normal parental lines: i.: C57BL/6J-congenic Fxnem2Lutzy (homozygous for floxed exon 2) ii.: C57BL/6-congenic Pvalbtm1(cre)Arbr Fxnem2.1Lutzy (heterozygous for both PV-Cre knock-in and frataxin global KO). These breeders produce the affected Fxnflox/null::PV-Cre genotype at a 1/4 frequency.

HUSBANDRY
Breeding Considerations
CD BioScience will be maintained by breeding the following phenotypically-normal mice: i. C57BL/6-congenic Fxnnull::PV-Cre (heterozygous for the frataxin global knockout allele and heterozygous for the Pvalbtm1(cre)Arbr allele) ii. C57BL/6J-congenic Fxnem2Lutzy (homozygous for the frataxin floxed exon 2 allele). Offspring from these breeding units will generate the affected mutant animals (Fxnflox/null::PV-Cre) at a frequency of 1/4. The genotype of Fxnflox/null::PV-Cre mice is compound heterozygous at the frataxin locus on chromosome 19 (Fxnflox/null ; floxed exon 2 allele on one homologous chromosome and a global knockout allele on the other homologous chromosome) and heterozygous for the Pvalbtm1(cre)Arbr allele. Due to the early-onset ataxic phenotype of such mice, we will only distribute Fxnflox/null::PV-Cre animals at 4-7 weeks of age. Of note, parvalbumin is expressed in sperm. However, characterization of Pvalbtm1(cre)Arbr mice performed by the Genetic Resource Science group reports no Cre expression in spermatids, and Pvalbtm1(cre)Arbr did not exhibit germline recombination in multiple crosses from different labs.
