Parp1loxP is a floxed allele with loxP sites flanking exon 4 of the poly (ADP-ribose) polymerase family, member 1 locus. Cre recombinase-mediated removal of the floxed sequence creates a PARP-1 knock-out allele. Parp1loxP mice may be useful in studying poly(ADP-ribosyl)ation (PARylation) in a variety of nuclear processes (including transcription, RNA processing and DNA repair).

The L1L2_Bact_P cassette was inserted at position 180578707 of Chromosome 1 upstream of the critical exon(s) (Build GRCm38). The cassette is composed of an FRT site followed by lacZ sequence and a loxP site. This first loxP site is followed by neomycin resistance gene under the control of the human beta-actin promoter, SV40 polyA, a second FRT site and a second loxP site. A third loxP site is inserted downstream of the targeted exon(s) at position 180579648. The critical exon(s) is/are thus flanked by loxP sites. A "conditional ready" (floxed) allele was Created by flp recombinase expression in mice carrying this allele to remove the lacZ sequence and neo selection cassette, leaving loxP sites flanking the critical exon(s).

Breeding Considerations |
Mice heterozygous or homozygous for this floxed allele are viable and fertile with normal breeding, and no reported gross physical or behavioral abnormalities. When maintaining a live colony, heterozygous mice may be bred together, to wildtype mice from the colony or to C57BL/6N inbred mice. Alternatively, homozygous mice may be bred together.
