The targeting vector contains an FRT flanked neomycin selection cassette and a loxP site inserted downstream of exon 5 of the targeted gene, and another loxP site was inserted upstream of exon 4. Flp-mediated recombination removed the FRT flanked neo cassette leaving exons 4 through 5 floxed.
