Molecular Note
A targeting construct was designed to insert a Cre Recombinase gene and a zsGreen (enhanced green fluorescence protein) just upstream of the 3'UTR of the angiotensin II receptor, type 1a (Agtr1a) gene. Viral P2A oligopeptides, from porcine teschovirus-1, were inserted upstream and downstream of the Cre gene to enable ribosomal skipping. A FRT flanked neomycin resistance (neo) cassette was placed downstream of the 3'UTR. This construct was electroporated into C57BL/6J embryonic stem (ES) cells. Correctly targeted ES cells were injected into blastocysts and the resulting chimeric mice were bred to FLP (FLPo) recombinase expressing mice to delete the neo cassette.
