A loxP site was introduced to the 5' UTR (50 bp upstream of the ATG start codon), and three repeats of the SV40 early terminator were placed in the 3' UTR (300 bp downstream of the stop codon) along with a second loxP site, the human placental alkaline phosphatase (AP) gene, and an FRT flanked neomycin resistance gene. The targeting vector was introduced to 129-derived embryonic stem cells. The neomycin cassette was excised with a germline FLP recombinase.
