NK1R-CreER knock-in/knock-out mice have the endogenous tachykinin receptor 1 promoter/enhancer sequences directing expression of tamoxifen-inducible Cre recombinase. These mice are a Cre-lox tool allowing inducible Cre recombination in Tacr-expressing cells/tissues (including neurons of the amygdala, hypothalamus, cortex, and spinal cord), and may be useful in studying Substance P signaling, inflammatory responses, and pain.

Molecular Note
The coding region of Tacr1 in exon2 was replaced by a Cre/ERT2 cassette and a loxP-flanked neo selection cassette by homologous recombination in ES cells. Correctly targeted ES cell clones were injected into C57BL/6 blastocysts. The neo-cassette was deleted by Cre-mediated excision.

Breeding Considerations
Both heterozygous and homozygous mice are viable and fertile with normal breeding, and no reported gross phenotypic or behavioral abnormalities. When maintaining a live colony, heterozygous mice may be bred together, to wildtype mice from the colony or to C57BL/6NJ inbred mice. Alternatively, homozygous mice may be bred together.
