A loxP site was inserted 300 bp upstream of the translation start site and a second loxP site was inserted along with an Frt flanked neo cassette immediately 3' of the coding region. The neo cassette was removed by in vivo Flp-mediated recombination leaving the entire coding region flanked with loxP sites.
