The RC::FPSit dual-recombinase intersectional allele has a frt-flanked STOP and loxP-flanked STOP that prevent transcription of synaptophysin-EGFP fusion protein and tdTomato. Exposure to FLP and Cre recombinase results in strong EGFP expression enriched at the synapse/synaptic vesicle (axonal boutons [terminal and en passant]) of the neuron subtype(s) defined by the overlap of FLP/Cre expression.

Site of Expression
After removal of both STOP cassettes by FLP- and Cre-recombination, fluorescent protein expression is observed in those cells where FLP and Cre expression overlaps - strong EGFP expression is enriched at the synapse/synaptic vesicle (axonal boutons [terminal and en passant]), whereas weak tdTomato expression is observed. EGFP expression is also observed in the soma and neurites to a certain degree.

Molecular Note
The targeting construct includes from 5' to 3' a CMV-IE enhancer/chicken beta-actin/rabbit beta-globin hybrid promoter, an FRT flanked STOP cassette, a loxP-flanked STOP cassette, a synaptophysin-GFP fusion gene (described below), IRES, a tdTomato gene, a WPRE, and a rabbit beta-globin polyA sequence. The synaptophysin-GFP fusion protein is a full-length mouse synaptophysin gene fused to a mutant enhanced green fluorescent protein (mut4EGFP; harboring the V163A/S175G codon changes for inCreased ability to fold properly at 37 degrees C).
