Atm floxed mice (also known as FX[M]) have loxP sites flanking exon 4 (encoding the N-terminal region) of the Atm gene. Exposure to Cre recombinase removes the floxed sequence - creating a null allele. Global knock-out mice generated from this floxed allele exhibit many aspects of ataxia-telangiectasia (A-T), including altered immunophenotype and behavioral defects, but without the increased incidence of lymphomas observed in many other models of A-T.

Molecular Note
Two loxP sites were inserted into the introns surrounding to exon 4. Flp-mediated recombination removed the FRT flanked neomycin resistance cassette inserted into intron 3. Cre-mediated recombination removed exon 4. This allele is predicted to cause a frameshift mutation through the splicing of exon 3 to exon 5. Western blot analysis confirmed the absence of protein expression in fibroblasts.

Breeding Considerations
Both heterozygous and homozygous mice are viable and fertile with normal breeding, and no reported gross phenotypic or behavioral abnormalities. When maintaining a live colony, heterozygous mice may be bred together or to wildtype mice from the colony. Alternatively, homozygous mice may be bred together.
