129S-Gt(ROSA)26Sortm3(CAG-luc)Tyj/J

Cat. No.: CEMM-07250147

A loxP-flanked puromycin resistance/STOP cassette followed by a fusion of firefly luciferase and synthetic antigen peptide SIYRYYGL were introduced to the Gt(ROSA)26Sor (gene trap ROSA 26, Philippe Soriano) locus. After Cre activity, the recombined locus has high expression of Luciferase-SIY fusion protein. This mutant mouse strain may be useful in studies of immune response to a self-antigen or over-expressed tumor-associated antigen when used in combination with tumor-prone models.

Site of Expression
When these mutant mice are bred to mice that express Cre recombinase, the resulting offspring will overexpress the luciferase-SIY fusion protein in the Cre-expressing tissues.

Molecular Note
A cassette containing a CAG promoter, a floxed STOP followed by a luciferase gene and sequence encoding the antigen SIYRYYGL (SIY) was inserted into the locus. Cre-mediated recombination is necessary to allow for expression of luciferase/SIYRYYGL under the control of the CAG promoter.
