Dctn1LoxP/ mice have loxP sites flanking exons 2-4 of the dynactin 1 gene. Cre recombinase-mediated removal of the floxed sequence results in continued expression of the p135 subunit, but complete loss of p150Glued subunit expression. These mice may be useful in studying the dynactin macromolecular complex in motor neuron disease and other neurodegenerative disorders.

A targeting vector was designed to insert a loxP-FRT neo-FRT cassette upstream of exon 2, and a loxP site downstream of exon 4 of the dynactin 1 locus (Dctn1) on chromosome 6. Flp-mediated recombination removed the selection cassette.

Breeding Considerations |
Mice homozygous for the floxed allele are viable and fertile, with normal fertility and no gross phenotypic or behavioral abnormalities. When maintaining our live colony, heterozygous mice may be bred together, to wildtype mice from the colony or to C57BL/6J inbred mice. Alternatively, homozygous mice may be bred together.
