B6(Cg)-Rhot1tm2.1Jmsu/J

Cat. No.: CEMM-07250847

Common Name: Miro1F
Exon 2 of the mouse Rhot1 (Miro1) gene is flanked by loxP sites in this Miro1F conditional mutant strain. Cre-mediated excision of the floxed region results in a knock-out allele useful in studies of mitochondria and motor neuron disease.

Molecular Note
A loxP site, FRT flanked neomycin resistance gene, exon 2, and a loxP site replaced exon 2. The FRT flanked neomycin resistance gene was removed via Flp-mediated recombination leaving exon 2 flanked by loxP sites.

Breeding Considerations
Heterozygous and homozygous floxed mice are viable and fertile. The donating laboratory notes that new litters are found on day 19.5 of gestation (when the day the plug was found is counted as 0.5). If late embryos are needed they have to be harvested on E18.5. Also, expression of Cre recombinase early in development instead of selectively in a specific tissue of interest will sometimes yield mosaic animals identifiable by genotyping (presence of more alleles than expected). The ectopic expression of Cre recombinase in the germline/early embryo can be problematic for this strain, since homozygous knockout animals are not viable, reducing effective litter sizes.
