The targeting construct is designed to contain a loxP site upstream of exon 3, an EYFP reporter fused to the 3' end of exon 4, a polyA, and an FRT flanked neomycin cassette followed by a second loxP site. Flp-mediated recombination removed the FRT flanked neo cassette.
