When these UBC-Cre-ERT2 mice are bred with mice containing a loxP-flanked sequence of interest, tamoxifen-inducible, Cre-mediated recombination will result in deletion of the floxed sequences in widespread cells/tissues. In Goodwin et al. 2019 Genome Res. 29:494, it was discovered that the transgene integrated into chromosome 2 causing a 5 bp deletion in the NADPH dependent diflavin oxidoreductase 1 locus (Ndor1).

This Cre-ERT2 transgenic mouse line was generated via lentitransgenesis; a self-inactivating lentiviral vector was designed with the human ubiquitin C (UBC) promoter sequence upstream of a Cre-ERT2 fusion gene (Cre recombinase fused to a G400V/M543A/L544A triple mutation of the human estrogen receptor ligand binding domain). Cre-ERT2-expressing lentivirus were microinjected into the perivitelline space of one-cell zygotes (129SvEv;C57BL/6 mixed genetic background). Mice display tamoxifen-inducible Cre activity in all tissue types. Line 1 inserted into the gene at 25249816-25249821 (Build GRCm38/mm10) resulting in a 5 bp deletion.

When maintaining a live colony, transgenic mice can be bred to wildtype siblings or to (129S1/SvImJ). A homozygous phenotype is not yet characterized (Aug-2007).
