These mice also contain a nuclear localized lacZ sequence downstream of the floxed region. Molecular Note
A targeting vector contained LoxP sites flanking the major coding exon 2 of Hic1, a PGK-neo cassette flanked by FRT sites distal to exon 2 and upstream of the 3' LoxP site. An IRES-NLS-LacZ cassette distal to the 3' LoxP site (downstream of exon 2) was added to enable the efficient identification of Hic1 deleted cells. The male chimeras were bred with female Flippase-expressing mice for deletion of the PGK-neo cassette.
