The R26NZG targeting vector contains a loxP-flanked PGK-Neo-4x polyA STOP cassette, FRT-flanked a nuclear localized β-galactosidase (nlslacZ)-3x polyA, and enhanced green fluorescent protein (EGFP)-1x polyA. These mice may be useful as a Cre mediated reporter or a FLP reporter when expressed in cells/tissues where the expression patterns of the individual promoters driving CRE recombinase and FLP recombinase overlap.

A vector containing a CAG promoter driving expression of a floxed neo/STOP cassette, FRT flanked lacZ gene with 3 polyadenylation sequences (secondary STOP cassette), and an EGFP gene was inserted into the locus. Expression of lacZ requires Cre mediated removal of the primary STOP cassette whereas expression of EGFP requires Cre mediated removal of the primary stop cassette and flp mediated removal of the secondary STOP cassette.
