C57BL/6N-Tg(Ppp1r2-cre)4127Nkza/J

Cat. No.: CEMM-07250234

Hemizygous Ppp1r2-cre mice are viable and fertile, harboring a BAC transgene encoding a Cre-recombinase gene under the control of the protein phosphatase 1, regulatory (inhibitor) subunit 2 (Ppp1r2) gene. These mice may be useful for studying the development and pathophysiology of schizophrenia-related disorders.

Site of Expression
When these mice are bred with mice containing loxP-flanked sequence, Cre-mediated recombination will result in deletion of the floxed sequences in the Cre-expressing GABAergic neurons of the offspring.

Molecular Note
BAC homologous recombination was used to insert Cre cDNA (fused with a nuclear localization signal (NLS) sequence) immediately upstream of the initiation codon of the Ppp1r2 gene in BAC RPCI-24-322B4. The Cre cDNA was followed by the bovin growth hormone-derived polyadenylation signal. In one clone, a 12-kb deletion introduced around the initial methionine codon of the apolipoprotein D (Apod) coding sequence, located 50 kb upstream of Ppp1r2, was identified. This clone was used to prevent overexpression of Apod in the brain of transgenic mice.

Breeding Considerations
When maintaining a live colony, hemizygous mice may be bred to wildtype (non-carriers) from the colony, or to C57BL/6NJ.
