Ai215 (also called Ai215 (TICL-stGtACR2-HA-ICL-GCaMP7s-SS)) is a Cre-dependent optogenetic and fluorescent calcium-indicator tool allele designed with a CAG promoter, loxP-STOP-loxP and stGtACR2 coding sequence followed by a CAG promoter, lox2272-STOP-lox2272 and GCaMP7s coding sequence - all inserted into the Igs7 locus (TIGRE). Exposure to Cre recombinase results in expression of stGtACR2 (soma-targeted variant of the red-shifted, light-gated anion channelrhodopsin GtACR2; neuronal silencing) and/or GCaMP7s fluorescence.

GtACR2 is derived from the Guillardia theta anion channelrhodopsin 2 gene.

Breeding Considerations |
Heterozygous mice are viable and fertile with no reported gross physical or behavioral abnormalities. To date, it has not been attempted to make this strain homozygous. When maintaining a live colony, heterozygous mice may be bred together, to wildtype mice from the colony or to C57BL/6J inbred mice.
