The entire coding region of the mouse Arc (activity regulated cytoskeletal-associated protein) gene is flanked by loxP sites in this Arclox conditional mutant strain. Cre-mediated excision of the floxed region in exon 1 creates a knock-out allele. Inactivation of Arc, involved in memory consolidation, prevents establishment of a strong conditional taste aversion that can develop as a consequence of food poisoning or chemotherapy treatment.

A loxP site was introduced to the promoter region and a loxP-FRT neomycin-FRT cassette was placed in the first intron downstream of the first exon. Flp-mediated recombination removed the FRT flanked neo cassette leaving the coding region floxed.
